  • The Effect of Green Tea Extractive Epigallocatechin Gallate on the Angiogenesis of HepG2 Transplanted Tumor in Nude Mice

    目的 通过复制人肝癌细胞株HepG2裸鼠皮下移植瘤模型,观察绿茶提取物表没食子儿茶素没食子酸酯(EGCG)干预对HepG2移植瘤新生血管生成的影响。 方法 瘤体接种复制HepG2移植瘤模型,荷瘤裸鼠20只随机分组,实验组给予EGCG溶液每日20 mg/(kg·只),腹腔注射3周,对照组给予等量灭菌注射用水3周,末次用药24 h,后处死裸鼠,剥离移植瘤。常规病理切片观察移植瘤组织结构;逆转录-聚合酶链式反应和免疫组织化学法检测移植瘤缺氧诱导因子-1α(HIF-1α)、血管内皮生长因子(VEGF)mRNA及蛋白表达,并通过检测CD34表达计数瘤组织微血管密度(MVD)。 结果 组织病理学观察实验组移植瘤见大量坏死区,瘤体内血管数量明显少于对照组;实验组HIF-1α、VEGF mRNA及蛋白表达水平比对照组均明显下调(P<0.05),实验组MVD比对照组明显下降(P<0.05)。 结论 EGCG可抑制荷瘤裸鼠HepG2移植瘤新生血管生成。

  • CHANGES OF EXPRESSION Fas OF THE ADRIAMYCIN-RESISTANCE HEPATIC CARCINOMA CELLS (HepG2 CELL LINES)

    Objective To investigate the effect of the drug-resistance characteristic of neoplasm cell on the expression of Fas during the chemical medi-cure.Methods The adriamycin-resistance hepatic carcinoma cells (HepG2 cell lines) were estabilished by cell biology. Changes of expression of the HepG2 cell lines was determined by immunohistochemistry. Results When the HepG2 cell lines were not induced by adriamycin, the expression of Fas of them was weak and Fas mainly existed in cell membrane. When induced by adriamycin, the expression was enhanced and Fas mainly existed in cytochylema. Simultaneously, the death rate of the cell lines changed. The death rate of the drug-resistance cell lines in 0.1 μg/ml ADM was almost as same as that of non-drug-resistance cell lines without ADM (P>0.05) and was significantly different from that of non-drug-resistance cell lines in 0.1 μg/ml ADM (P<0.05). Conclusion Changes of the expression of Fas may be one of the drug-resistance mechanisms of carcinoma cell.

  • SUPPRESSION OF HUMAN HEPATOCELLULAR CARCINOMA CELL GROWTH OF RETINOBLASTOMA SUSCEPTIBILITY GENG

    The Chinese human hepatocellular carcinoma cell SMMC7721 has been analysed by flow cytometry, Southern blotting and Western blotting. The results indicated that SMMC7721 cell is a hypoploid cell with a 0.81 DNA index, and that SMMC7721 cell has internal deletion in the 5'-end of its Rb gene and has no Rb gene product (Rb protein). The normal Rb cDNA has been inserted into a retrovirus vector DOL and introduced into SMMC7721 cells by electrporation transfection technique.About 75% of transfected SMMC7721 cells have been killed by Rb gene product. The remain 25% cells are alive as exogenous Rb gene has been mutationally inactivated. (Chin J Ocul Fundus Dis,1994,10:21-24)

  • ADVANCES IN EPIDEMIOLOGIC STUDY OF PRIMARY LIVER CANCER

    ObjectiveTo study the epidemiologic characteristics of primary liver cancer (PLC). MethodsThe literatures about regional distribution and etiologic epidemiology of PLC were reviewed. Results PLC was mainly distributed on caostland in the south-east of China. The main cause of PLC was hepatitis B virus, aflatoxin and contamination of drinking water. Otherwise, PLS was also related with lack of some trace element, sex horemones, genealogy cause and so on.Conclusion The genesis of PLC was by multiple factors.

  • Efficacy and safety of laparoscopic versus open hepatectomy for hepatocellular carcinoma: a meta-analysis

    Objective To systematically review the efficacy and safety of laparoscopic hepatectomy (LH) and open hepatectomy (OH) for patients with hepatocellular carcinoma (HCC). Methods PubMed, EMbase, The Cochrane Library, CBM, WanFang Data, CNKI databases were electronically searched to collect the case-control studies about LH vs. OH for patients with HCC from inception to December, 2015. Two reviewers independently screened literature, extracted data and assessed the risk of bias of included studies, then meta-analysis was performed by using RevMan 5.3 software. Results A total of 28 studies involving 1 908 patients were included. The results of meta-analysis showed that: the LH group was superior to OH group on complications (OR=0.35, 95%CI 0.26 to 0.48, P<0.000 01), hospital stay (MD=–4.18, 95%CI (–5.08, –3.29),P<0.000 01), and five years overall survival rate (OR=1.65, 95%CI 1.23 to 2.19,P=0.000 7) and disease-free survival rate (OR=1.51, 95%CI 1.12 to 2.03, P=0.006). However, no significant differences were found in one year and three years overall survival rate, disease-free survival rate, and postoperative recurrence rate. Conclusion Current evidence shows that the LH is superior to OH for the treatment of HCC, and may be amenable to surgery because of its safety and longtime efficacy. Due to limited quality and quantity of the included studies, more high quality studies are needed to verify above conclusion.

  • Experience of Diagnosis and Treatment of 53 Cases of Intraperitoneal Hemorrhage Due to Spontaneous Rupture of Primary Carcinoma of Liver

    目的 探讨原发性肝癌自发性破裂腹腔内出血的临床特点和手术治疗方法。方法 回顾性分析我院1998~2007年收治的53例经手术证实的原发性肝癌自发性破裂腹腔内出血患者的临床资料,结合文献复习进行讨论。结果 53例患者均以突发性腹痛为首发症状,有前期症状者33例,无症状者20例; 有外伤史者(左上腹或季肋区)6例; 有腹膜炎体征46例,腹腔穿刺抽出不凝血41例,合并休克29例。术前误诊16例。行急诊手术48例,施行肝切除33例。术后肝功能不全34例,其中3例死于肝功能衰竭。术后生存3 d~78个月,平均34.6个月。结论 原发性肝癌自发性破裂腹腔内出血多以腹痛为首发,常以血性腹膜炎表现为特点,术前误诊率较高; 手术方式根据病变位置、大小,肿瘤是否局限、转移和肝硬变程度决定。术后近期死亡原因主要是肝功能衰竭,远期死亡原因主要是复发转移。

  • CORRELATION BETWEEN SYNDECAN-1 PROTEIN EXPRESSION AND MALIGNANT PHENOTYPES OF HEPATOCELLULAR CARCINOMA

    Objective To investigate the relationship between syndecan-1 protein and malignant phenotypes of hepatocellular carcinoma (HCC). Methods forty-seven formalin-fixed sections were obtained. The syndecan-1 was measured with immunohistochemistry assay (ABC method). Results Among 47 HCC tissues, 28 (59.6%) show negative staining, the syndecan-1 negative rate in HCC with poor differentiation was higher than those with good differentiation (78.3% vs 41.7%, P<0.05), and negative rate in large HCC was higher than in small HCC (81.0% vs 42.3%, P<0.01), and negative rate in HCC with the presence of tumor-cells in blood was greater than in those without tumor-cells in blood (84.2% vs 42.9%, P<0.01). No correlation was found between syndecan-1 expression and serum AFP level and Child class. Conclusion Syndecan-1 expression is correlated with the growth, differentiation, invasiveness, metastasis and progression of HCC. It is possible that syndecan-1 is a negative regulator of these malignant phenotypes of HCC, can be regarded as suppressive gene.

  • INTERNAL RADIOTHERAPY OF LIVER CANCER

    Objective To study the development of internal radiotherapy for liver cancer and the relationship between effects and radiation doses. Methods Literature about internal radiotherapy of liver cancer were collected and reviewed. Results The rational selection of radioactive microsphere,the appropriate control of radiation dosage and the path of internal radiotherapy are crucial in improving the therapy effects and decreasing the complications. Conclusion The two-stage operation of liver cancer which is on the base of combining chemotherapy, radiotherapy and immunotherapy is the way to go of liver cancer therapy.

  • GUIDED CHEMOEMBOLIZATION THERAPY FOR MODERATE AND ADVANCED CARCINOMA OF LIVER USING ADRIAMYCIN LIPIODOL EMULSION (A REPORT OF 884 CASES)

    We have performed guided chemoembolization on 84 patients of moderate and advanced carcinoma of liver using adriamycin lipiodol emulsion (A/L) since 1986. Result showed that the rate of improvement of symptoms was 86.1%, in 75% cases the AFP were decreased and in 79.2% the size of tumor were reduced. The mean survival time was 10.3 months which was much higher than that of the control group (5.6 months,Plt;0.001). THe survival rates of 1/2,1,2,3 year were 89.3%,43.4%,13.5% and 3.8% respctively that were significantly higher than those of the control group (51.2%, 11.5%,0) (Plt;0.01). Three patients underwent secondary resection after using A/L chemoembolization ans gelatin spinge central embolization with a longer survival rate. This may be a good method of treatment to the nonresectable liver cancers and may also be an easy way for postoperative observation.

  • Construction and Expression of The Recombinant of Hepatocellular Carcinoma-Targeting Adenovirus Containing r-Caspase-3 Gene

    Objective To construct the recombinant of hepatocellular carcinoma-targeting adenovirus containing r-Caspase-3 gene and provide the gene therapic strategy for hepatocellular carcinoma. Methods The pAdTrack-EAFP-PALB was constructed and the r-Caspase-3 gene was subcloned into the vector. The linearized shuttle plasmid was homogenously recombined with AdEasy-1 in BJ5183 cells. The candidate clone was analyzed by restriction endonuclease digestion and sequencing, and then pAdEasy-EAFP-PALB/r-Caspase-3 vector was digested with PacⅠand transfected into AD293 cells for packaging and amplifying, recombinant virus was constructed successfully. Infection titer and efficiency of recombinant virus were monitored by green fluorescent protein (GFP) expression. The expression of r-Caspase-3 in infected HepG2 cells was detected by RT-PCR and Western blot. The apoptosis of HepG2 cells was detected by SRB dyeing method. Results Shuttle vector pAdTrack-EAFP-PALB/r-Caspase-3 was correct after identification by restriction endonuclease analysis and sequencing. By PCR and PacⅠ restriction endonuclease analysis, the homologous recombinant of pAdEasy-EAFP-PALB/r-Caspase-3 was successful. The expression of GFP was observed when linearized pAdEasy-EAFP-PALB/r-Caspase-3 was transfected into AD293 cells. AD293 cells could be infected repeatedly by recombinant adenovirus. The expression of r-Caspase-3 gene on HepG2 cells was detected by RT-PCR and Western-blot methods respectively, which confirmed that the Ad-EAFP-PALB/r-Caspase-3 was constructed successfully. The specificity of Ad-EAFP-PALB/r-caspase-3 which targeting induced hepatocellular carcinoma cells was founded by SRB dyeing test. Conclusion The Recombinant of hepatocellular carcinoma-targeting adenovirus containing r-Caspase-3 gene was constructed successfully and which established the foundation of r-Caspase-3 gene therapy in future research to hepatocellular carcinoma.
