Objective  To observe the gene expression of Nrf2 in cisplatin-resistant lung adenocarcinoma cancer cells, and explore regulatory effects of metformin on it. Methods  The A549 cells and drug-resistant A549/DDP cells were cultured and intervened by different concentrations of metformin. The proliferation inhibition rate and apoptosis rate of A549 and A549/DDP cells were detected by cell proliferation experiment and flow cytometry. The gene and protein expressions of drug resistance related genes (Nrf2, GSH, and MRP-1) were detected by real time PCR and Western blot. Results  The gene expressions of Nrf2, GSH, and MRP-1 in A549/DDP cells were significantly higher than those in A549 cells. Metformin could inhibit the gene expression of Nrf2 and its downstream genes (GSH and MRP-1), and further inhibit cell proliferation and induce apoptosis of A549 and A549/DDP cells. Conclusion  The expressions of Nrf2 and its downstream genes differs in different lung adenocarcinoma cell lines which can be significantly suppressed by metformin.
